T4 polynucleotide kinase
DESCRIPTION
Isolated from a recombinant E. coli strain this enzyme catalyzes the transfer of the terminal phosphate group of ATP to 5'-OH ends of DNA. T4 polynucleotide kinase is suited for labelling of 5'-OH ends of DNA with a 32P-ATP.
APLICATION
End-labeling DNA or RNA for probes and DNA sequencing
Addition of 5´-phosphates to oligonucleotides to allow subsequent ligation
Removal of 3´-phosphoryl groups
CONCENTRATION
10 units/µl
UNIT DEFINITION
One unit is the amount of enzyme that incorporates 1 nmol of acid-insoluble 32P in 30 minutes at 37ºC.
STORAGE BUFFER
10 mM K-phosphate buffer pH 7.0; 100 mM NaCl; 0.5 mM EDTA; 1 mM DTT; 0.01% Tween 20; 50% glycerol (v/v)
STORAGE TEMPERATURE
Store T4 polynucleotide kinase below 0ºC, preferably at -20ºC, in a constant temperature freezer.

350 mM Tris-HCl pH 7.6; 50 mM MgCl2; 25 mM DTT
